Anthelmintic activity of aminoalcohol and diamine derivatives against the gastrointestinal nematode Teladorsagia circumcincta

Gastrointestinal nematodes (GIN) infections are a serious problem in livestock production due to the great economic losses they cause. Their control is increasingly difficult because of rapid development drug resistance and limited number available drugs. Therefore, this study evaluated 18 aminoalcohol 16 diamine derivatives against eggs, first third stage larvae from susceptible resistant isolate Teladorsagia circumcincta collected sheep. The effectiveness vitro anthelmintic activity compounds was using three different procedures: Egg Hatch Test (EHT), Larval Mortality (LMT) Migration Inhibition (LMIT). Those with activities higher than 90 % initial screening at 50 ?M were selected determine their half maximal effective concentration (EC50). In parallel, cytotoxicity assays conducted on Caco2 HepG2 cell lines calculate Selectivity Indexes (SI) for each compound. 30 presented best results preventing egg hatching, displaying lowest EC50 value (1.01 ± 0.04 ?M) all tested highest SI (21.21 vs. Caco-2 cells). For LMIT, 34 showed efficacy, values 2.67 0.08 3.02 0.09 parasite, respectively.
